چکیده

The effect of bleomycin on the molecular integrity of SV40 DNAwas determined under various conditions by taking advantage of the fact that a single scission produced in such a superhelical form of DNAcould be clearly manifested by analysis of samples with centrifugation in alkaline sucrose gradients. Conditions were found where the reaction velocity, the rate of disappearance of the characteristic form of DNA,depended on the concentration of bleomycin as well as on the reaction time. In contrast to general enzymatic reactions, bleomycin activity was greater at 0°C than at 37°C. The optimum pH was 9.1 while no activity was observed below pH 6 or above pH 13. Additional DNA,irrespective of its source, inhibited the reaction, while RNAhad no effect. Cu++ion at 1 mMalmost completely inhibited the reaction while 2-mercaptoethanol at 1 mMstimulated the reaction by about 20fold. No sulfhydryl compound was included in our standard assay system. EDTAat 10 mM showed no effect.

Sperm DNA damage in mice irradiated with various doses of X-rays alone or in combination with actinomycin D or bleomycin sulfate: an in vivo study

Background: DNA damage in male germ cells due to exposure to environmental and manmade physico-chemical genotoxic agents is considered as the main cause of male infertility. The aim of this study was to evaluate the effects of combined modalities (radiotherapy and chemotherapy) routinely used for cancer treatment on mouse sperm chromatin in vivo. Materials and Methods: Forty-eight mice were div...

DNA strand scission in vivo and in vitro by auromomycin.

The mechanism of action of auromomycin, a new tumor-inhibitory antibiotic, was studied in a growing culture of mouse lymphoblastoma L5178Y cells and with isolated viral DNA. Auromomycin prevented growth of L5178Y cells completely and irreversibly at antibiotic concentrations higher than 0.03 microgram/ml. DNA synthesis was preferentially inhibited by the antibiotic, whereas RNA and protein synt...

The production of hydroxyl radical by tallysomycin and copper(II).

Tallysomycin is an antitumor antibiotic that is structurally related to bleomycin. The mechanism of action of tallysomycin appears to be similar to that of bleomycin. Bleomycin induces strand scission of purified isolated DNA [ l--5]. Bleomycin has also been shown to produce breakage of intracellular DNA [5,6]. Iron(H) appears to be associated with the mechanism of action of bleomycin. The foll...

Effects of DMA Superhelical Changes Induced by Ethidium Bromide on the DMA-degrading Activity of Two Antitumor Antibiotics, Bleomycin and Phleomycin1

The effects of changes in the conformational state of DNA on the single-strand and double-strand breakage activity of two antitumor antibiotics, bleomycin (BLM) A2 and phleomycin Di, have been studied by the gel electrophoretic analysis of the drugdegraded PM2 phage superhelical DNA pretreated with an inter calating agent, ethidium bromide (EB). Both the single-strand and double-strand breakage...

Intracellular DNA strand scission and growth inhibition of Ehrlich ascites tumor cells by bleomycins.

A comparison of the intracellular DNA strand scission activities of the antitumor drug bleomycin, three of its metal complexes, demethyl bleomycin A2, and iron-containing, redox-inactivated bleomycin in Ehrlich ascites tumor cells was performed by means of the alkaline elution technique. This comparison was aided by use of CoCl2 to eliminate or minimize post-cell lysis strand scission by bleomy...
